Epigallocatechin-3-Gallate Suppresses the Expression of TNF-α-Induced MMP-1 <i>via</i> MAPK/ERK Signaling Pathways in Human Dermal Fibroblasts

Deeper wrinkles and loss of elasticity are one the skin-aging symptoms. Collagen breakdown by matrix metalloproteinase-1 (MMP-1), which is induced reactive oxygen species (ROS) pro-inflammatory cytokines, has been known to be responsible for these Therefore, much attention paid chemicals suppress MMP-1 activity. Epigallocatechin-3-gallate (EGCG), catechin rich in green tea, reported show antioxidant protect skin from various stimuli such as UV chemicals. In this study, we evaluated inhibitory effect EGCG on gene expression secretion tumor necrosis factor-α (TNF-α)-treated human dermal fibroblast cells (Hs68 cells). Pre-treatment with (10 20 µM) suppressed TNF-α-induced secretion. also reduced phosphorylation extracellular signal regulated kinase (ERK) significantly but not that p38 activation c-Jun N-terminal (JNK). Besides, showed mitogen-activated protein (MEK) Src upstream proteins ERK pathway. Based results, might have potential activity slow down through inhibition collagen breakdown, remains elucidated.
